Inhibition of the p42/p44 MAPK activity by PD 98059 suppresses MAPK nuclear translocation. CCL39 ΔRaf-1:ER cells were grown for 2 d on glass coverslides and serum-deprived for 24 h. Before fixation and p42/p44 MAPK immunolabeling, cells were treated as follows: (A) control cells were left unstimulated; (B) cells were stimulated for 3 h with 10% FCS; (C) cells were stimulated for 3 h with 10% FCS in the presence of 30 μM MEK inhibitor PD 98059; (D) cells were incubated 3 h in presence of 30 μM PD 98059; (E) cells were stimulated for 3 h with 1 μM estradiol; (F) cells were stimulated for 3 h with 1 μM estradiol in the presence of 30 μM PD 98059.
