Figure 7.
15-Epi-LXA4 attenuates lung inflammation and prolongs survival of Escherichia coli–infected mice. One hour after intraperitoneal injection of 2 × 108 live E. coli, BALB/c mice were treated intravenously with vehicle or 15-epi-LXA4 (200 μg/kg). Mice were killed at 6 hours post E. coli and (A) lung tissue myeloperoxidase activity, (B) BAL fluid neutrophil count, (C) the percentage of annexin-V–positive (apoptotic) neutrophils, (D) neutrophil caspase-3 activity, and (E) lung dry-to-wet weight ratio were determined. Data are means ± SEM (n = 6 mice per group). *P < 0.05, **P < 0.01. RFU = relative fluorescence units. (F) Lung tissue sections from naive mice (control), mice challenged with E. coli and treated with 15-epi-LXA4 or vehicle. Hematoxylin and eosin stain; scale bars: 100 μm. (G) Kaplan-Meier survival plots for mice challenged intraperitoneally with 109 live E. coli and treated with 15-epi-LXA4 or vehicle (n = 10 per group). P = 0.0136 by the log-rank test.
